Hi first do the basic checks, like hosted application or web page is working in the hosted machine means base machine if that is working fine then is so easy to fix the issue if not open IIS logs and start analysis , any bugs or error in the web page take help of devloper and fix it

To do the database configuration please follow the MS article or web page is working fine in base mation but not able to access in internal network or from external network cross check DNs entry and routing and firewall entries
