3. Alcohol or Acetone Decolorization: The key step in Gram staining is the decolorization step. The slide is rinsed with either alcohol or acetone. This step differentiates between two types of bacteria:

- Gram-Positive Bacteria: These bacteria have a thick peptidoglycan layer in their cell walls, which traps the crystal violet-iodine complex. The alcohol or acetone does not effectively penetrate this thick layer, so the stain remains trapped, and these bacteria retain their purple color.

- Gram-Negative Bacteria: These bacteria have a thinner peptidoglycan layer and an outer membrane, which is dissolved by the alcohol or acetone. This allows the crystal violet-iodine complex to be washed out of the cells, causing them to lose their purple color.

4. Counterstaining: To complete the staining process, a red counterstain (usually safranin) is applied to the slide. Gram-negative bacteria, which lost the purple color during decolorization, take up the safranin stain and appear red under the microscope.

The result of Gram staining is that bacteria are classified into two broad groups:

- Gram-Positive Bacteria: These bacteria appear purple under the microscope due to the retention of the crystal violet-iodine complex in their thick cell walls.
